The breakthroughs in recombinant protein production permitted BioConcept Ltd. the ability to produce and supply a new type of cell culture media. MAM-PF ®, known as Mammalian Artificial Media, is a line of media tailor made to satisfy the needs of researchers and producers. MAM-PF® series media is Animal Component Free (ACF) media and is in accordance with the strict quality guidelines EMA/410/01. This Cell Culture Media is protein-free and protein hydrolysates free. The special media is chemically defined and is used for high cell density cultivation of a variety of cell lines such as CHO (Chinese Hamster Ovary) cells or BHK (Baby Hamster Kidney) cells and the high-level expression of recombinant proteins. BioConcept holds a certificate for every single component used in the MAM-PF® media series to guarantee an untainted and exceptional final product.
This cell culture media is formulated without L-glutamine to avoid problems associated with L-glutamine degradation including ammonia accumulation. The Mammalian Free Media can be obtained with a minimal amount of Phenol Red or without Phenol Red. The serum-free culture represents an advantage over serum supplemented culture as it facilitates purification and downstream processing of expressed products. Compared to other commercially available serum-free media, the MAM-PF® does not contain various undefined proteins or protein hydrolysates.
CHOSI cells cultured in the Animal Component Free Media, MAM-PF77®, show the fastest growth along with a ~100% and ~40% higher cell density compared to media by Suppliers S and T. When looking at the graph, this corresponds with the final product titers at the end of the fed-batch. All the cultures in the display were fed with FMS3 in the same feed regime. The higher viability in the stationary phase shows that the glycosylation in Cell Culture Media, MAMPF77®, was superior.
When mixing the CHO feed mix FMS3 to MAM-PF77®, a cell density of over 100 mio. Cells per liter can be reached. A titer of over 350 mg/L of highly glycosylated folliclestimulating hormone (FSH) is possible to achieve within 14 days of stirring in a bioreactor tank. This demonstrates that the cell culture media is a very high-quality product. As determined during the purification process and as shown on the graph, 45% of the product displays an isoform-pattern, low aggregates, and low oxidized forms. Thus meaning, MAM-PF77® can be used to produce quality FSH that fulfils its Ph.Eur. requirements.
The continuous innovation and development of the MAM-PF® cell culture media has led to an addition called MAM-PF77® cell culture medium and CHO Feed Mixes FMS3 and FMU. MAM-PF® media can increase productivity of Ipilimumab (monoclonal antibody) by up to 5g/L, which is shown in the graph over a 14-day trial. The new expression system is also viable in fed-batch and perfusion systems.
